people have said this many times, but it's an easy, fast way to display your work. straight to the point, foolproof.
although I plan on making my full portfolio multiple pages, I still think one pagers, when executed well, are just as, if not more effective as a multiple.. It all depends what you are trying to sell yourself on. Someone who is mainly a print designer doesn't need a flashy website.. If you are a flash programmer though, you'd better come out with some heavy artillery. (obvious)

i get a lot of compliments from less "websavvy" people who just want a quick overview of my work. what is so hard about scrolling down? it couldn't be easier, no?
and no, i am not trying to look cool a la "hey i can make a website", i can actually build pretty good sites and more complicated but that is simply not the point here my friend.
key is:
simplicity, accessability and to leave a quick and good impression with potential clients.

the only possible negatives I can think of for one page portfolios is the limitation on number of images shown and the fact that the proximity of other images may influence the one you're looking at. These may actually be positives, though.
