Dont even think about anything else than the MUSEO DE ANTROPOLOGIA E HISTORIA, dude seriously, if you into contemporary art, go to JUMEX MUSEUM in Polanco, a very impresive art collection.
If you have time go eat at EL PARNITA or EL BELMONDO Both in Colonia Roma, really nice eats at a very decent price.
Night time drinks and shit, try to do to FELIX or ROMITA across the street, Both in Colonia Roma
If you venture into Centro, go to SALON CORONA, classic shit, good Octopus Tacos.
Boom there you go...

Are you alone or with someone that knows the area?
Keep in mind, Mexico City is HUGE and traffic is insane, so getting around isn't that easy and very time consuming. So it will all depend on where your staying. I would stick to taxi's. Ask your hotel about which are the safe ones to take.My personal pick would be Coyoacan and visit Frida Kahlo's home, La casa Azul. Then head over to the "Centro" (downtown) and have some beer and amazing al pastor tacos at "Salon Corona". http://www.saloncorona.com.mx
If you want to check out another museum, check out "Museo de Antropologia" some amazing stuff in there and in the same area you will find "El Castillo de Chapultepec" yes a castle that is in the middle of Chapultepec park. Those museums will show you a few sides of Mexico's history.
- ^ looks like ambush had it covered with salon corona and the museum. lolsea_sea

Try Pozole at Casa de Toño there are a bunch I would recommend the one in Santa Maria de la Rivera (chain restaurant and not fancy at all but still badass.

one night? El polankito (Polanco, Miguel Hidalgo, Mexico City, Mexico) tell any taxi driver take you there. Its a whole street with dive bars, restaurants, clubs. you find somethings there for sure. Its also full of foreigners. English speaking is a plus.
