OS X tips/tricks
Window views: the column view is (in my humble opinion) the most efficient file browsing available in OS X. Start typing to select a folder, and use arrow keys for a pure keyboard Finder experience.
Window sidebar: a great place to keep custom Smart Searches and folders you access regularly or temporarily. You can even toss in network folders and drives. Good stuff.
Smart Searches: man, these are helpful. I'm not going to get all into why, but I suggest checking them out. In your sidebar you'll see a Search For twistie and some preset searches. Right-click on one and Show Search Criteria to see how they're created. The level of filter detail is astonishing and powerful. Press Command-F (Find) to initiate a new search (or Command-Spacebar to initiate a Spotlight search). The real-time search results and customizability in OS X put other OSs to shame, in my opinion, of course.
In the Finder, go to Preferences and explore your options. You can create a really clean desktop by hiding connected drive icons (they're already accessible in Finder windows) from the desktop and snapping the icons to grids and stuff like that.
Command-Tab works just like Ctrl-` in XP (last Windows I've used, so I can't say anything about Vista or 7), but a little nicer. The consistent key modifiers in the OS and applications is really handy too, N for new (window or file) and E for eject in the finder, which is handy for quick HD removal. I use Command-delete a lot to send files to the trash without using a mouse. Stuff like that.
If I think about anymore, I'll let you know.

This was mentioned before but I'd just like to add,
The new version of Transmit is really good. It's clear they put a lot of effort into tweaking every little aspect of the user experience. Let's hope the new Coda is as much improved.

When you install the OS, go to Advanced and check out the extras you can add. Hopefully it's not to late to suggest this.
