If you're there on a Sunday - do the East end.
Start with a breakfast @ Premises cafe, a walk on Columbia Road Flower Market, then down to Brick lane, do the markets, have a pint in the vibe bar. Job Done.

I just moved to London from New York and have found some great places. I think the key to London is to have a few options picked out wherever you go (a restaurant, a pub, a park etc.) because there are some horrible and overpriced places you don't want to go to...

Its kind of hidden in an old school bike shed, but the menu changes everyday and food is always fresh and delicious. Its BYO with a £5 corkage fee. Its only open Mon-Fri 11-3, so you might want to make a res.Vinoteca (Clerkenwell)

Super cute pub. Always good food. Nice atmosphere.Definitely second the vote for Columbia Road Market. Its amazing. Its only on Sundays and finishes around 2:30, so don't go too late.
Broadway Market is also really great. I LOVE the samosas at the stand called Gujarati Rasoi. Its at the top of the market, close to the park. Seriously amazing.
Borough Market is amazing as well. Especially love the chorizo and rocket sandwiches from Brindissa.

tate, go shopping in covent garden & seven dials, get lashed in camden and see a gig, take a walk down the kings road, go celeb spotting in primrose hill, get robbed at knifepoint in stoke newington.

Fantastic food and very unique style of restaurant. Go there your first night, and have a look through Wardour Street (where it is) to scope out another meal as there are some nice restaurants along there that are reasonably priced.
Go to Spitalfields/Brick Lane on a Sunday, if you don't mind getting up vaguely early on a Sunday. Get there by going to Aldgate East (Brick Lane end) or Liverpool Street Station (Spitalfields end).

For a good panoramic view of London, on a clear day, take a walk up to Primrose Hill (Chalk Farm tube station - Northern Line). Followed by lunch & drinks in one of the local bars. You coud walk down the hill and check-out London zoo after.
What Gordy says for Sundays is a winner.
