If you're there on a Sunday - do the East end.
Start with a breakfast @ Premises cafe, a walk on Columbia Road Flower Market, then down to Brick lane, do the markets, have a pint in the vibe bar. Job Done.
As it goes, I did a map for it for a mate who's going there.

Fantastic food and very unique style of restaurant. Go there your first night, and have a look through Wardour Street (where it is) to scope out another meal as there are some nice restaurants along there that are reasonably priced.
Go to Spitalfields/Brick Lane on a Sunday, if you don't mind getting up vaguely early on a Sunday. Get there by going to Aldgate East (Brick Lane end) or Liverpool Street Station (Spitalfields end).
Tate Modern maybe?

For a good panoramic view of London, on a clear day, take a walk up to Primrose Hill (Chalk Farm tube station - Northern Line). Followed by lunch & drinks in one of the local bars. You coud walk down the hill and check-out London zoo after.

How long are you there?
I did 10 days on mine...5 in London, 5 in Paris. Easily doable...about a 3 hour train ride.
You might want to take a tour to the old Roman city of Bath....see the town & Stonehenge...it's about 100 miles north of London.
