I've had my wee MBP down to 10 gigs or thereabouts for several months now. It's been chugging along happily, but gets bogged down.
Regrettably, I've even had it down to roughly 80 megs a couple of times and had to purge all kinds of shit and only been able to get it back up to about 10 gigs.
The culprits are many, some work files, some MP3s, etc. Mostly though, the serious issue is the gigs worth of pics I've imported from my DSLR. I've got them backed up on a Lacie 1TB external hard drive, but I've heard horror stories of people having HDs blow up and losing all kinds of data so I avoid relying on it too heavily.
What reliable means can you guys recommend to lighten the load while keeping files safe and accessible?

When I'm done with a project our group of files I zip them up and keep them in a folder called Project Archive and delete the originals.

Funny that you avoid backing up onto an external HD, especially when the one inside your computer isn't much different (and probably more prone to failure due to transport). Your best bet is to just buy HDs and do what version3 said.

Multiple HD backups. Time Machine the OS drive and daily stuff.
i also archive my work into annual folders and rip off to DVD at end of year and store in a spindle at my folks place incase my house burns down or gets royally looted.
someone steals your computer they'll most likely grab any externals beside it.

keep one backup offsite. multiple backups are useless if you have a fire or flood.

i'll tell you what it is, it's that damn spotlight that's indexing your hd and slowing things down for all your other apps.

Main storage for quick access or current project files I used the mac HDD.
I backup to two external WD NAS Drives (MyBook World Editions), manually after I finish a project as well as having Time Machine do its thing. I have one drive for personal stuff and another for work related stuff. NAS drives are great with a mbp because you can access it anywhere within WiFi and even access it remotely with the software that comes with it.
Then I make an additional backup online to my web server after I finish every project just in case a plane lands on my roof. This takes a while to do since you have to upload it. I'll usually do this EOD so it can do its thing overnight. This also comes in handy when you want access to files that you don't have locally and when you're working remotely when your external HDD is inaccessible.
Every few months I'll burn some DVD's just so I have another so-called safe backup solution. Just in case a plane crashes into the servers location.

Hey, get a different external hard drive. Lacie 1Tb are bad for dying... at least the power cords are. Mine just quit, won't mount and just spins. Done some reading tho and it seems the power cords fail before the drives do so I'm hoping (and praying) I can't get a new cord and everything will be fine.
Good thing I put all my work on that one drive. And good thing it's most of my old graphic work. Not my music. Not my samples or family pictures. It's just old work... I don't need it... unless, wait, I need to make a portfolio... DAMMIT.
Multiple backups is really good advice.
