The best system i ever used was OSX server with AIT back-ups.
Server never (and that's never) went down and the back up was as fast as flock.
If you can afford it get yourself a decent Xserve set up, you won't regret it.

i work at a game studio and we back up all our artists and programmers 3 times a week to tape. old school man, but it's the only thing the can hold it all and be consistent in keeping it somewhat reliable.

For the server, if you can afford it, get an Xserve. Best file server on the planet. App servers, get an IBM or a Dell.
For back-ups, tape lasts long, and is very durable, more so than DVD, and hold more too. But dvd is still an excellent option.
