    Normally for large format its 300dpi at 25%. So I'd get it as good as you can at 25%. Scale it to 300dpi in PS and make sure you have 'resample image' unchecked. Then if you need to scale it up further you can, add some noise to sharpen it up and get rid of artefact from scaling.

    Also remember that if its a poster its not really meant to be viewed up close, the print is usually much coarser so you have a little leeway on it.

    The best advice though, is talk to your printer. See what size and res they need it at and work to that.
