Scripting language is a matter of preference. Most here would likely recommend php (I would too) but there are others; dotnet, webobjects, etc, etc.
You'll likely want to go with an existing cart rather than building a new one from scratch. Use filter — there are about a million of discussions about e-com carts.
Depending on scale you may be looking for a team rather than a single dev-guy.
