What is the best way to inline typography in Illustrator?

My problem is to make the inner line smooth in the rounded corners of for example D, U and S.

Best way might be to do a negative offset path on the letterform using the rounded option. It will need to be converted to outlines first.

Yep, turn font into outlines ( CMD-SHIFT-O ) and then offset path.
Object > Path > Offset Path

The best way to inline a font is to use the pen tool. If you can't draw a simple curve in illustrator its time you learned!
Offset paths is not going to give you what you want. You can get a thinner letter this way, but not the single inline like you want per the example.
