in my experience make it as quick and easy as possible for them to read it. when responding via email i would always put a short cover letter and resume in the body, attaching a pdf version of the resume for them to print for further review. also, up at the top of the email i would put a few of my latest links. they always click the links.
then they like what they see and read my short cover letter. then download the resume. then i got a job.
