Definitely depends on what's in your comp. Multiprocessing can be a double edged sword depending on if you have enough RAM to back it up. It can slow down a render if it gets bottlenecked. A good rule of thumb is to only allow multiprocessing if you can give each core 2GB of RAM on it's own. Since CS5 is 64-bit, it has the ability to take advantage of all of the RAM in your machine, instead of just 4GB per app. I rarely run into memory buffer issues in CS5, and I used to hit them constantly in CS4.
I like to run on a 3 disk setup, as was previously mentioned:
1) System drive with the OS and app
2) Scratch drive (striped RAID is ideal). This is where your cache should live
3) Media driveI also like to keep my autosave folder on my system drive (not on the RAID drive or where I keep my .aep files) as a safeguard.
I don't really think the size of the disk cache matters (as far as being too big). I give it 50GB or more.
