Send a letter along with each package. Do some homework up front to find out exactly whom you should send packages to, rather than simply "Dear Marketing Director". It also wouldn't hurt to look into whether they ever outsource design work at all. If they have an existing relationship with an outside designer, no matter; but if 100% of the work is done in-house, you'll have wasted your time, their time, and the cost of a package for every un-researched mailer you send.
Make your portfolio front and center. Don't try to be too clever or obtuse or designery. This is for potential clients, which I see as something totally different than when you send a package to creative directors who are potential employers. Potential clients need to see immediately what it is you do and what specifically you can do for them. They don't need to wade through a conceptual essay. Trust me, I've learned this one the hard way.
