Family Guy has more laughs per minute than the Simpsons.. the jokes and punch-lines are more succinct and the humour is far more wicked / cutting. Imo, Family Guy's format feels more like a vehicle for delivering jokes, whereas the Simpsons is more constrained to a traditional narrative.
It isn't all spitting, punching and farting; there's loads of depth to Family Guy and some of the references are very clever (sometimes too clever).
Simpsons would win for me; probably because I grew up with it. Simpsons is an institution - the whole universe that the show's created is amazing and there are so many quality characters.
