I have built a lot of my own furniture for home, and it can be quite time consuming and expensive.
Your looking at a good chunk of change for right tools and wood; not to mention the time it will take to build. Plus, there are so many extras to consider: clamps, screws, hardware, finishes and space to build.
Like Moural said, I would do a few sketches and get the sizes. I assume you would want to use plywood and face it wil some 1 x 2 pine or something to hide the plywood edge. Use a softwood for sure (excluding the top of the desk) as it is more forgiving when making joints.
Oh, make sure to use some good tools too - sharp saw blades and chisels.
