    yes - not having all your font managed by OSX FontBook should speed up your computer at startup etc.

    my separate drive/ partition suggestion is more about keeping your box clean and efficient.. font libraries tend to get huge (in terms of # of files) and are the files are prone to damage. its best to isolate them somewhere where they can be easily scanned and repaired on their own. plus when you optimize or repair your system disk there won't be 1000's of fonts to scan in the process, which can be hella slow.
