I design on 1024x768 at home (stupid no spanning on iBook), and 1280x1024 at school (17" LCD). I have a peecee with 1280x1024. My next will probably be either a widescreen 20" LCD or aa 17" iMac... widescreen is so killer for designing, and watching movies!
Always design for 800x600, or use a trick to detect resolution.
People are paying you to do design, so you've got an obligation to make their site available to everyone.
If its personal work, design for a jumbotron... but i will hit the back button before it finishes loading
