It all depends on YOUR particular audience... our site has way more users at 1600x1200 than 640x480....
...and someone on a 1600x1200 or larger screen is usually NOT running the browser full-screen... they've got a few browser windows over here... maybe some 3D modelling going on over there, a few shell windows... etc... don't assume you have their whole screen to play with...
