The Mall is an easy place to start Most of the Smithsonian's are located right there in addition to the National Gallery (not part of the Smithsonian ). Hirshorn and the Freer & Sackler Gallery on the Mall as well as the West and East Bldg of the National Gallery are must sees.
The National Bldg Museum F & 4th Street (via Judiciary Sq. Metro) is another nice museum (actually, the bldg. itself is really worth checking out).
Adams Morgan, Dupont Circle, or Georgetown are all neighbohoods worth visiting.
If you're real adventerous, head over to Capitol Hill and get a tour of the Jefferson Bldg (part of the Library of Congress) and I believe you can get a library card as well to 'check out' books. The Main Reading Room of the Jefferson Bldg. is very impressive. From there, you can walk over to the Capitol Bldg. and get a tour or you might still be able to walk around unguided (not sure how they do it now-a-day).
If you do go to Capitol Hill, head east down Pennsylvania Avenue to the the Greek restaurant that's about two blocks from the Capitol. Get a Chicken Souvlaki. You'll thank me later.
Definitely take the Metro wherever you go. Relatively cheap and easy public tranportation system. The stations have a really unique architecture to them...worth photographing as long as some security cop doesn't think you're a terrorist.
