Almost everything we print is from photopolymer. We produce a plate from a piece imagesetter film, so any digital file you have can go to press. We often gang stuff up on our press sheets for jobs like identity systems and invitations with many different pieces. (invite, map, RSVP, save the date, etc on the same sheet) That saves a lot in set up costs. Most letterpress shops can not print that large. My best advise is to contact us earlier in your design process. Good production strategy is key to quality letterpress.
Here is a poster size letterpress print I designed for a show called Poster Offensive that coincided with the RNC convention here in the Twin Cites:

Have you thought about engraving? It's the opposite effect of letterpress (raised lettering), and it uses a copper or steel die which is crisper. Letterpress is awesome, don't get me wrong, but engraving has more finesse to it. These guys are the best:
- Best in U.S., at any rate. Dickson's is good too, but much more $$$.gramme

Going with letterpress has always made me think twice due to lack of knowledge and ignorance on the cost.
