I am going to be working on a 30 second video that will be shown on a very large screen in times square... this is my first time doing something this large... any pointers?

yeah dude, make sure you're working with the tech guys at the display company. this stuff is tricky. i once did a video similar to this and they wanted the quicktime to be something small like 240x180. i was skeptical, but it turned out looking great.

if it is LED screens you only need to know the resolution, which is, depending on the density of the LEDS (different systems out there) is usually not so large.

last summer a friend and i did some visuals for a huge screen (and with huge i seriously mean huge, about 8-10 meters high) on a large festival and it turned out great on normal DV-Pal format. (even got cut of a little, so we could have done with smaller!)
