It's just a stupidly broad question, digdre - if you've never been before, pick up a tourist guide and go with the suggestions therein - they'll more than suffice for a mere weekend. Start in Piccadilly, work your way out from there on foot. Find a hotel on the Piccadilly line - you don't need to be too central if you're willing to start off each day with a 15 minute tube journey. If you do want to stay central, search for hotels in Holborn, though I'm not sure how tight your budget is. Try lastminute.com for cheap deals.
You can't go wrong with the various Tate museums ('musea'?) and a walk along the Thames from, say, Parliament to London Bridge - there're a lot of sites on either side of the river along that route.
If you want more design-centric stuff, dig through the filter here, as there have been plenty of threads on the subject - though be careful not to bore the shit out of your Dad!
